Antiproliferative Activity of Violaxanthin Isolated from Bioguided Fractionation of Dunaliella tertiolecta Extracts ArchiMer
Pasquet, Virginie; Morisset, Perrine; Ihammouine, Said; Chepied, Amandine; Aumailley, Lucie; Berard, Jean-baptiste; Serive, Benoit; Kaas, Raymond; Lanneluc, Isabelle; Thiery, Valerie; Lafferriere, Mathieu; Piot, Jean-marie; Patrice, Thierry; Cadoret, Jean-paul; Picot, Laurent.
Dunaliella tertiolecta (DT) was chemically investigated to isolate molecules inhibiting cancer cell proliferation and inducing apoptosis in vitro. The potency to inhibit cell growth was used for the bio-guided fractionation and isolation of active compounds using chromatographic techniques. The DT dichloromethane extract exhibited a strong anti-proliferative activity on MCF-7 and LNCaP cells, and was further fractionated and sub-fractionated by RP-HPLC. High resolution mass spectrometry and spectrophotometric analysis unequivocally identified violaxanthin as the most antiproliferative molecule present in DT DCM extract. Violaxanthin purified from DT induced MCF-7 dose- dependent growth inhibition in continuous and discontinuous treatments, at...

Changes of the Total Lipid and Omega-3 Fatty Acid Contents in two Microalgae Dunaliella Salina and Chlorella Vulgaris Under Salt Stress BABT
Rismani,Sharare; Shariati,Mansour.
ABSTRACT Effect of salt stress on biomass, cell number, contents of total lipid, omega-3 fatty acids, including ALA (Alpha Linolenic Acid), EPA (Eicosapentaenoic Acid) and DHA (Docosahexaenoic Acid) and their biosynthetic pathway intermediates (palmitic acid, stearic acid, oleic acid and linoleic acid) of two microalgae Dunaliella salina and Chlorella vulgaris were investigated. Dilution stress from 1.5 to 0.5 M NaCl and salt stress from 1.5 to 3 M NaCl were incorporated into the D. salina medium. Salt stress of 200 mM NaCl was also applied to C. vulgaris culture. Results indicated that increasing salt concentration resulted in the reduced growth rate of C. vulgaris and substantial increase of the total lipid content in both species. Proper growth rate of...

Effects of nitrogen limitation on Dunaliella sp.–Alteromonas sp. interactions: from mutualistic to competitive relationships. ArchiMer
Le Chevanton, Myriam; Garnier, Matthieu; Lukomska, Ewa; Schreiber, Nathalie; Cadoret, Jean-paul; Saint-jean, Bruno; Bougaran, Gael.
Interactions between photosynthetic and non-photosynthetic microorganisms play an essential role in natural aquatic environments and the contribution of bacteria and microalgae to the nitrogen cycle can lead to both competitive and mutualistic relationships. Nitrogen is considered to be, with phosphorus and iron, one of the main limiting nutrients for primary production in the oceans and its availability experiences large temporal and geographical variations. For these reasons, it is important to understand how competitive and mutualistic interactions between photosynthetic and heterotrophic microorganisms are impacted by nitrogen limitation. In a previous study performed in batch cultures, the addition of a selected bacterial strain of Alteromonas sp....

Interactions microalgues-bactéries en système expérimental bispécifique : effets sur la croissance de Dunaliella sp. ArchiMer
Le Chevanton, Myriam.
The use of algae-bacteria mixed culture has been suggested to improve performance of industrial microalgae culture. The present study aims to investigate impact of selected bacteria on Dunaliella sp. growth. 97 bacteria strains were isolated from a variety of marine microalgae cultures. The diversity of strains revealed members of α-and γ-proteobacteria, Cytophaga-Flavobacterium-Bacteroides and grampositive. A two-phase approach was then implemented to select bacteria that improve Dunaliella sp. growth, using an original high-throughput experimental device. Although a majority of bacteria induced performance reduction for Dunaliella sp., some resulted in significant improvements. In nitrogenlimited batch culture, the Alteromonas sp. bacteria demonstrated a...

Producción de biomasa y carotenoides de Dunaliella tertiolecta en medios limitados en nitrógeno Phyton
López-Elías,JA; Fimbres-Olivarría,D; Medina-Juárez,LA; Miranda-Baeza,A; Martínez-Córdova,LR; Molina-Quijada,DMA.

Proteomic Analysis of the Chlorophyta Dunaliella New Strain AL-1 Revealed Global Changes of Metabolism during High Carotenoid Production ArchiMer
Ben Amor, Faten; Elleuch, Fatma; Ben Hlima, Hajer; Garnier, Matthieu; Saint-jean, Bruno; Barkallah, Mohamed; Pichon, Chantal; Abdelkafi, Slim; Fendri, Imen.
The green microalgae Dunaliella genus is known for the production of high added value molecules. In this study, strain AL-1 was isolated from the Sebkha of Sidi El Hani (Sousse, Tunisia). This isolate was identified both morphologically and genetically via 18S rRNA gene sequence as a member of the genus Dunaliella. Strain AL-1 was found to be closely related to Dunaliella salina, Dunaliella quartolecta and Dunaliella polymorpha with more than 97% similarity. Response surface methodology was used to maximize carotenoid production by strain AL-1 by optimizing its growth conditions. The highest carotenoid content was obtained at salinity: 51, light intensity: 189.89 μmol photons·m−2·s−1, and nitrogen: 60 mg·L−1. Proteomic profiling, using two-dimensional gel...

Screening and selection of growth-promoting bacteria for Dunaliella cultures ArchiMer
Le Chevanton, Myriam; Garnier, Matthieu; Bougaran, Gael; Schreiber, Nathalie; Lukomska, Ewa; Berard, Jean-baptiste; Fouilland, E.; Bernard, Olivier; Cadoret, Jean-paul.
Previous studies have demonstrated that bacteria influence microalgal metabolism, suggesting that the selection and characterization of growth-promoting bacteria should offer a new strategy for improving industrial algal cultivation. In the present study, 48 cultivable bacteria were isolated from marine microalgae species and identified using 16S rRNA phylogenetic analysis. The recovered bacteria were found to be members of the α- and γ-Proteobacteria, Cytophaga–Flavobacterium–Bacteroides (CFB) and gram-positive monophyletic clusters. To address the effect of these bacteria on the growth of Dunaliella sp. individually, an experimental high-throughput tool was developed to simultaneously compare replicated associations. A two-step approach was used to...
